Fluorinated functionalisation and sp3-hybridised carbon atoms are two of the most exciting areas being explored in medicinal chemistry today. Research has shown that three-dimensional ring structures such as cyclopropane and cyclobutane offer the possibility of increased solubility and the opportunity of accessing biological targets with more globular shapes. Furthermore with a proven track record of successful progression through clinical testing, as demonstrated in the leukotriene C4 synthase inhibitors - WO2016177845 (AstraZeneca) and the orexin receptor antagonists - US20120095031 (EIsai) it is no surprise that sp3 hybridised cycloalkyls are increasing in popularity. The presence of fluorine has also been well documented, particularly in influencing the metabolic stability and physico-chemical properties of compounds. Whilst most studies have focussed on the lowering of pKa values on the basic nitrogen in heterocyclic, an increased number are looking into fluorinated cycloalkyls as an alternative.
